Inaugurato nel 1951 ed intitolato a Mecenate, illustre cittadino aretino ed importante personaggio della Roma augustea, il Museo ha sede nel convento dei Montolivetani di San Bernardo, la cui costruzione fu iniziata nel XIV sec. su parte delle strutture dell'anfiteatro romano; di quest'ultimo sono infatti visibili, al piano inferiore, le volte dell'ambulacro.
